Why this requirement deserves technical definition
Glucose, glutamine, pyruvate and buffering differences matter when specifying a cell-culture medium. The practical objective is to define the complete workflow well enough that product selection, sourcing and substitution decisions preserve the scientific intent.
1. DMEM is a family of formulations, not one universal recipe
Dulbecco’s Modified Eagle Medium is available in multiple formulations that differ in glucose, glutamine, pyruvate, bicarbonate, phenol red and other components. “DMEM” alone is therefore not a complete procurement specification when a validated cell-culture protocol depends on a particular formulation.
2. Cell line and protocol define what must remain fixed
The required formulation should be taken from the established protocol, cell-bank documentation or validated development work. High-glucose and low-glucose variants, for example, are not interchangeable merely because they carry the same general medium name.
3. Supplements and serum should be specified separately
FBS, antibiotics, non-essential amino acids, growth factors and other supplements may be added by the laboratory rather than supplied in the basal medium. Separating the basal formulation from the supplementation recipe avoids ambiguity and makes alternatives easier to evaluate.
4. Manufacturer references are technical context, not an authorization claim
